چکیده

Gordon and Jackson (1) found that synthetic monomethyltryptophan was capable of stimulating growth in rats subsisting on a diet deficient in tryptophan. Chin (2) isolated kynurenine and kynurenic acid from the urine of rabbits after the parenteral administration of monomethylL-tryptophan (MMT). On the other hand, recent studies have shown that the albino rat, pig, horse, cotton-rat, and man can convert dietary tryptophan to niacin or its methylated derivative, W-methyhricotinamide (W-Me) (3). In the present paper the urinary excretion of niacin and W-Me by rats after the ingestion of MMT was studied.

Change of tryptophan-niacin metabolism in D-galactosamine-induced liver injury in rat.

The change of tryptophan-niacin metabolism in D-galactosamine (D-galN) injected rats was investigated. Rats fed with niacin-free diets containing 40% casein for 11 days were injected with D-galN (0.8 g/kg body weight). The urinary excretions of nicotinamide and its metabolites, and the activity of liver alpha-amino-beta-carboxymuconate-epsilon-semialdehyde decarboxylase (ACMSD) (EC 4.1.1.45), a...

Niacin biosynthesis in the developing chick embryo.

In 1945, Krehl et al. (1) observed that tryptophan and nicotinic acid were mutually interchangeable, within limits, in supporting the growth of rats. Since that time considerable evidence has accumulated which demonstrates the biosynthesis of niacin from tryptophan in the rat and other animals. Studies with Neurospora and mammals have demonstrated that the pathway from tryptophan goes through k...

Studies on the Biological Activity of Nicotinylalanine, an Analogue of ICynurenine*

Considerable evidence has been obtained that 3-hydroxyanthranilic acid and quinolinic acid can be converted to niacin in the intact rat (2-9). The usually low yield of niacin from these compounds suggested that another pathway from tryptophan to niacin might exist. Thus, kynurenine or hydroxykynurenine might be converted to niacin through an intermediate such as y-(3-pyridyl)-y-oxo-ar-aminobuty...

The influence of vitamin B6 on the formation of liver pyridine nucleotides.

It has been shown that, when tryptophan is administered to rats, an increased excretion of niacin and some of its derivatives occurs (l-4). Isotope studies by Heidelberger et al. (5, 6) have given further evidence that niacin can be derived from tryptophan in the animal body. Vitamin Bs has been implicated in this transformation, since in a pyridoxine deficiency the excretion of an abnormal met...

The Enzymatic Conversion of Quinolinate to Nicotinic Acid Mononucleotide in Mammalian Liver.

It has been known for many years that tryptophan is converted to niacin by mammals (1) and Neurospora (2) and that 3-hydroxyanthranilate is an intermediate in this conversion (3). The steps between 3-hydrosyanthranilate and niacin have, homever, remained obscure until recently. A very active soluble oxidase present in mammalian kidney and liver converts 3-hydroxyanthranilate to the unstable acy...
